La adquisición de tolerancia a la desecación en semillas ortodoxas es una estrategia adaptativa que no presentan las semillas del tipo recalcitrantes y por consiguiente son sensibles a la desecación. El grado de tolerancia o sensibilidad a la desecación se ve afectada por varios aspectos de origen genético, fisiológico, bioquímico y anatómico; por consiguiente existen diferencias entre genotipos y entre individuos dentro del genotipo de la misma especie. In Colombia, the cocoa pod husk (CPH) is expected to reach 2 100 000 t year-1 in 2021 which is usually burned or left over for decomposing outdoors at the plantations without any environmental control. Therefore, this study evaluated the energetic potential of CPH obtained after the initial processing of this fruit (Theobroma cacao L.). Three biological materials were analyzed: clone CCN-51 (CPH1), clone ICS-39 (CPH2) and a hybrid (CPH3), which present high yield and number of fruits per tree. ABSTRACT This study was conducted to evaluate the effect of substituting Tifton 85 hay (Cynodon sp. cv. Tifton 85) with levels of cocoa (Theobroma cacao) byproduct (0, 8, 16, and 24%, diet dry matter basis) in the diet of 16 Holstein × Gir heifers with an average body weight of 363.0±27.7 kg on their nutrient intake, digestibility, and protein balance. The experimental period was 17 days, and the sample collection period was five days. There was no effect of cocoa byproduct on nutrient intake. En el presente trabajo, se propuso realizar una determinación bioquímica de la actividad enzimática de aminopeptidasa (tcAPE) durante los procesos de germinación y fermentación de la semilla de T. cacao L., así como amplificar los extremos 3’ de fragmentos de cDNA de los posibles genes codificantes. El trabajo inició con la determinación de la actividad enzimática de tcAPE. O objetivo do trabalho foi verificar o efeito do ácido indolbutírico (AIB) no enraizamento de estacas do cacaueiro. O delineamento experimental foi o inteiramente casualizado, em esquema fatorial 3 x 5 x 2, envolvendo 3 clones (Cepec 2008, CCN 51 e TSH 1188), 5 concentrações de ácido indolbutírico AIB (0; 1.000; 3.000; 6.000 e 9.000 mg kg-1), duas épocas do ano (verão e inverno),, cinco repetições e 10 estacas por parcela. Genetic distance and its association with heterosis in cacao BABT

The efficiency of cacao breeding program can be increased by choosing superior crosses to be made between divergent clones. We assessed the genetic distance among five clones with RAPD data (genetic distance - GD) and with yield component data (Mahalanobis distance - MD). The clones were evaluated in a diallel, during five years, for five yield components. A total of 130 RAPD bands were scored. GD and MD were used to determine the correlation between genetic distances among clones and the performance of their hybrids. The correlation between GD and MD was 0.67 (P=0.03). ABSTRACT Levels of Zn in tropical soils profoundly influences growth and nutrition of tree crops. Research was undertaken to assess the effect of soil Zn on growth and nutrition of clonal cacao tree seedlings of PH 16. Three acidic Oxisol soils differing in texture were used with nine doses of Zn (0, 1, 2, 4, 8, 16, 32, 48, and 64 mg dm-3). Rooted clonal seedlings were grown in plastic pot with 11 dm-3 of the soils at varying Zn levels for 240 days. At harvest growth (dry matter mass of leaves, stems, shoots, roots, and total) and nutrient concentrations were determined. The clonal cacao seedlings showed differences for production of dry matter mass and foliar nutrient concentrations for P, K, Ca, Mg, Mn, Fe, Zn, and Cu. There was Zn toxicity in all soils.

Purificación y caracterización bioquímica de la aminopeptidasa (tcAPE) de la semilla de cacao (Theobroma cacao L.). Colegio de Postgraduados

La aminopeptidasa tcAPE fue parcialmente purificada de semillas germinadas de cacao (Theobroma cacao L.) de diez días de germinación, en cuatro pasos de purificación 1) Homogenización y extracción de la enzima, 2) Precipitación con sulfato de amonio al 60% de saturación, 3) Concentración con el sistema de ultrafiltración Amicon® Millipore empleando una membrana de 10 kDa de corte de peso molecular y 4) Cromatografía de intercambio iónico. Purificación y caracterización bioquímica de la carboxipeptidasa (tcCP) de la semilla de cacao (Theobroma cacao L.). Colegio de Postgraduados

La carboxipeptidasa tcCP de semillas de Theobroma cacao L., fue purificada 11.5 veces con un redimiendo de 45.5% con tres pasos de purificación que incluyen: homogenización y extracción de la fuente de enzima, precipitación con sulfato de amonio al 40% de saturación y cromatografía de intercambio iónico. El peso molecular de la enzima se estimo por electroforesis en gel de poliacrilamida al 12%, en condiciones desnaturalizantes (SDS-PAGE), con tinción con plata. La tcCP es un monómero con peso molecular de 30.77 kDa, el cual hidroliza péptidos a temperatura de 40-50ºC y pH 7.0. El sustrato Bz-Tyr-pNA fue hidrolizado a una mayor proporción a 10 mM (13.576 UAE/mg de proteína total) confirmando la presencia de actividad carboxipeptidasa (CP) (E.C. 3.4.16)....

Agroclimatic zoning for agricultural sector and creation of land suitability maps by fuzzy logic contribute to such production expansion. ABSTRACT: The objective of this research was to establish soil patterns and foliar standards for clonal crops of cocoa CCN51 and PS1319 in the Atlantic regions within the northern Espírito Santo and southern Bahia States. Samples were collected from September to October 2015, in 45 high productive plots, being 23 plots of clone CCN51 and 22 of clone PS1319. Sufficiency ranges for soil and foliar standards were recommended for cocoa clones CCN51 and PS1319 planted in the Northern Espírito Santo and Southern Bahia states. O Estado de São Paulo é uma das regiões em que se cultiva o cacaueiro (Theobroma cacao L.) mais afastadas do equador, com características climáticas distintas daquelas registradas nas regiões tradicionais de cultivo. A manteiga de cacau apresenta características físico-químicas peculiares, estreitamente relacionadas aos teores dos ácidos graxos que a constituem, os quais são influenciados pelas condições climáticas, especialmente a temperatura do ar. Variation and its distribution in wild cacao populations from the Brazilian Amazon BABT

A sample of 64 progenies (320 cacao trees as a whole) from four Brazilian Amazon basins was collected and evaluated on the basis of 15 fruit and seed traits. Nested univariate analyses of variance showed significant variation across progenies and basins. However, most of the variability appeared to be due to among trees and basins differences. The multivariate analysis showed that the differentiation in cacao populations occurred among basins. Since cacao diversity was predominantly found in trees within basins and among basins, one should optimize the collecting process by taking as many trees as possible starting from few progenies and many river basins. ABSTRACTThe main objective of the present research was to calculate the water footprint of the Colombian cocoa (Theobroma cacao L.) production. The evaluation of crop water requirement and irrigation requirement were based on climate, soil and crop conditions in the country. The water requirement estimation was based on data from six municipalities selected for their representativeness of the highest yield, productivity and commercial dynamics of the country. The results show that the Water footprint reached 17,100 m3 t-1.
